Research overview

CJC-1295 without DAC is also called modified GRF (1-29). Its chain has 29 residues, a free amino terminus and an amidated carboxyl terminus. The four substitutions are D-alanine, glutamine, alanine and leucine at positions 2, 8, 15 and 27. The Jetté paper investigated maleimido derivatives and their albumin conjugates, not the isolated peptide without the lysine–maleimidopropionyl extension [1].

That study prepared three maleimido derivatives of hGRF(1-29) and conjugated them to serum albumin ex vivo. It examined attachment through the free thiol of albumin Cys34, stability against dipeptidylpeptidase-IV and growth hormone secretion in cultured rat anterior pituitary cells. The rat experiments included Western blot analysis of plasma to examine material associated with serum albumin. The selected albumin-binding construct was named CJC-1295; these experiments do not characterise the peptide lacking DAC [1].

The second paper investigated in vitro metabolism and detection in fortified urine of four synthetic GHRH analogues: sermorelin, tesamorelin, and CJC-1295 with and without the DAC extension. Researchers identified 19 major in vitro metabolites, then synthesised, purified and characterised them as reference materials. The analytical work used these metabolites together with parent peptides and the sermorelin fragment sermorelin(3-29)-NH2 to develop a liquid chromatography–tandem mass spectrometry method for detecting the peptides [2].
