Research overview

SNAP-8 is the octapeptide recorded as acetyl octapeptide-3, with the residue sequence Glu-Glu-Met-Gln-Arg-Arg-Ala-Asp. An acetyl group caps its amino terminus and an amide caps its carboxyl terminus. These terminal groups are part of the molecular identity rather than additional amino-acid residues. Acetyl octapeptide-3 was an ingredient in both cited patch studies. The sequence contains two glutamate and two arginine residues, with one methionine, one glutamine, one alanine and one aspartate [1][2].

The first paper reported a single-centre study of a hyaluronic acid microneedle patch containing several ingredients. Alongside acetyl octapeptide-3, the formulation contained arginine/lysine polypeptide, palmitoyl tripeptide-5, adenosine and seaweed extracts. The study used instrumental analysis to examine skin hydration, density and thickness. Its experimental material was the complete patch formulation, with several peptide and nonpeptide constituents assessed together [1].

The second paper reported a placebo-controlled evaluation of a dissolving microneedle patch made with a hyaluronic acid polymer backbone. Its additional ingredients included acetyl octapeptide-3, L-ascorbic acid 2-glucoside and sodium cyclic lysophosphatidic acid. The comparator patch contained hyaluronic acid alone. The authors described fabrication by droplet extension and assessed mechanical strength and transepidermal water loss. Neither cited study separated the contribution of SNAP-8 from those of the other ingredients [2].
